Will "let's do an exercise" also work or is this something of a compound verb in practice, where it always refers to exerciseS?

Yes there is;

Exercise (noun) --> Egzersiz --> "physical activity."

"Egzersizi yapalım." --> Let's do the exercise (physical)

"Egzersizleri yapalım." --> Let's do the exercises (physical)

In another comment in a past exercise, one of the native speakers said it can mean either a physical exercise or an exercise that you would do when studying.

Thanks for the response! :) I've had a chance to ask a native speaker in Turkey. He said it's definitely not used for physical exercise. For that one, we should use egzersiz yapmak.

In translation 'let's excercise' is okay, but i think 'do' is required when yapar is used, at least where duo is concerned
