I tried "zwei plus zwei gleich vier" and it was wrong. I thought that was the same as using "ist" or "gibt" in this context?

You're right! At least in simple statements like Zwei plus zwei gleich vier, you can use ist, gleich, or ist gleich. In fact, your answer is now accepted.

When using the word gleich in more complex statements, just be sure to use the dative case following gleich, as exemplified here:
Der Flächeninhalt eines Rechtecks ist gleich dem Produkt aus seinen beiden Seitenlängen. (The area of a rectangle is equal to the product of the lengths of its sides.)
