Er du den eldste would be fine. Not det.
If you referred to a car, is it the oldest = Er den den eldste, den being the masculine pronoun because car is a masculine noun, and you are close. And also er det den eldste where det is a determinator, maybe you are pointing at it, but usually that would be "is that the oldest" in English.
