I imagine that "trâu" refers mostly to water buffalo, yes? Can it also refer to North American plains bison?

Bison are known as bò rừng (forest bovines). You can also call them bò bison/bizon ("bison" bovines) and if you want to specify the continent of America then it's bò bison châu Mỹ or bò rừng châu Mỹ.
