"זה פסיק, לא נקודה."

Translation:It is a comma, not a period.

That "niqudah"...is it somehow related to "niqqud"?

Yes! Literally נקודה is a point of a dot, nikud is the action of "dotting".

I hear that he says "nekudáh", not "ni-".

But don't worry, I hear it is a very "comma" mistake to make! Hahaha! I'm sorry. Pity my relatives.

How are פסיק and פיסקה related?

Well, פָּסַק means "to stop, cease". A פְּסִיק "comma" makes an incision in a sentence, a פִּסְקָה "paragraph" in a text; in both cases you have a slight stop in your reading.
