No, there's no difference between them. There's nothing like ser and estar in Romanian.

it depends on how fast you're talking and on your style. so "nu este greu", "nu e greu" and "nu-i greu" are all the same.

There's no difference between "este" and "e". "e" is just a short form for "este". In some cases you'll see "i" instead of "e" as a kind of contraction written always with a hyphen "-i" for example: "Nu-i aici cu mine"
