Is preparar strictly prepare, as in making a sandwich for a buffet and having someone eat it a while later or could I say in a restaurant. Me pode preparar um sanduíche? if I am hungry and want her to make me one right away? Duo didn't allow make in this instance, so I was wondering.

Yes, you can use preparar for something to be made right away. "Você pode me preparar um sanduíche de frios?" would be perfectly suitable :)
