Because Άγιος is stressed on the 3d to last syllable followed by an unstressed word, it should get a second syllable "Ο άγιός μου"

Yes! It's either "Η άγια" (more modern) or "Η αγία" (older style) but you will hear both versions in Greece, the second one perhaps more common.
Ah - didn't know there were alternative pronunciations - thanks. However female saints are not currently accepted in either form.
