(Oops, sorry) "... is tallest" you need the "la" because más alta = taller, la más alta = tallest.

Ah, I see. Yes in English you can say, "she is tallest" or "she is the tallest", but in Spanish the la is always necessary.

As DanD_8 says, it's needed to form the phrase equivalent to "the tallest one." Without it, you would just be saying "my sister is taller."
