Does město sound like menesto to anyone else?

It is pronounced [mňesto]. See Tips and Notes about the pronunciation of mě and mně.

town-city Is there any difference?

City is bigger, I think it had to have a cathedral. In Czech we have just one word for both - město.

You can also say "velkoměsto", lit. "bigtown". The usage is slightly different than with city, as velkoměsto implies a positive evaluation. Also the size necessary for a place to be called velkoměsto differs depending on the speaker. I'd say minimum is 100.000 people, but it really depends on the kind of city life you can get there. Compare with maloměsto = a place whose inhabitants have very limited, conformist or even hypocritical views, used to show disapproval.
