Should it be "Kamu berdiri"? It currently says "Kau berdiri."
It's all correct, but usually "kamu" sounds friendlier than "kau". In formal sentences, "Anda" would be used.

Scenario: angry professor telling one of his students to stand up. Would he say just berdiri (stand up) without kamu or that is unnatural?
In your scenario, that sentence is considered a command, so yes, the professor won't need to say "kamu" anymore, it's like in English, "stand up!". But the sentence in this exercise here is not a command.
