is there any difference between "bangunan" and "gedung"?

Gedung = Building.
Bangunan = Construction, structure, building.

They are not exactly the same, or synonyms.
For example, a tower can be called "bangunan" , but it's not a "gedung".

If I use "memasuki", then the building is the object of the sentence and I can leave out the "ke"'right?
"Saya mau memasuki bangunan itu."

"Saya mau memasuki bangunan itu" is also grammatically correct, but it just doesn't feel natural.

"we build a building" = "kita bangun/membangun sebuah bangunan"
