Yes, it doesn't change the meaning. Adding it just puts more emphasis on that you only want one piece.

Doesn't yksi make the bread definite? Why is leipä in partitive in this case?

Yksi leipä = one bread (whole thing, or could be also one sandwich), whereas yksi pala leipää = one piece OF the bread.
