shouldn't this be "ela pensa sobre o livro". the word "no"in this sentence seems wrong

pensa em (or generally pensar em) is translated as "think about". There are other circumstances where "em" is translated as "about" also, and after pensar is definitely one of them. Now "em o" is contracted to "no", hence "think about the..." = "pensa no"

So...it tells me "no" is a new word....it gives me 4 definitions. None of them are "of".....?

"of" is not a literal translation for "no". The best literal translation for "no" (= em + o) here is "about the" (em = at/on/in/about. So the best literal translation in English is "She thinks about the book", which has the same meaning as "She thinks of the book".
