Even though "en" can mean in, at, on - in this type of sentence we usually say "at", "at my house", "at their house", "at his house", etc, especially in past tense.
"I was at her house yesterday"
"I am in my house right now."
But you can use either in/at in present tense...
"Where are you?" - "I am at Juan's house" (ahora)
"Where are you?" - "I am in my house" (ahora)
