" You have no sense of humour" is the correct phrase, it means you cannot understand a joke. The other use of humour is " in good humour", " in bad humour", or in an " odd humour" etc.
That is exactly correct. For the rest of the phrases you mention here the translation in Greek would be "διάθεση = mood" and not "humo(u)r" though.
